Super Kings' seam bowlers Mustafizur Rahman, Deepak Chahar, and Tushar Deshpande each took two wickets.
Earlier, New Zealand's Rachin Ravindra scored an explosive 46 off 20 balls, while captain Ruturaj Gaikwad also scored 46. Chennai posted 206-6.
Shivam Dube was the top scorer with 51 runs off 23 deliveries, boosting the total after Ravindra and Gaikwad gave the team a quick start with their opening stand of 62.
MS Dhoni, the old warhorse, surprised his fans by passing on the Chennai captaincy to Gaikwad ahead of the T20 tournament, even though he had led the team to their fifth IPL title last year.
Gaikwad, who led the team to victory over the Royal Challengers Bengaluru in the IPL opener last week, survived an early reprieve on one and made good use of the opportunity.
Ravindra, who is left-handed, remained defiant and hit three sixes and six fours.
Gujarat's spin bowler Rashid Khan prevented Ravindra from reaching his fifty when he had the batsman stumped. Gaikwad was caught behind off Australia's Spencer Johnson.
Dube reached his fifty in just 22 balls before falling in the 19th over to Rashid, who took two wickets but leaked 49 runs.
